چکیده

In a preceding paper (1956) the writers reported that pigmentation occurs in varyinig degree among strains of most species of the proposed but as yet undescribed new genus, Dekkeromyces. Hybrids may yield more pigment than either parent, thereby permitting easy selection of hybrids from a population consisting of both hybrids and parent species. This method of isolating hybrids requires one or both of the parents to produce a small or moderate amount of pigment, but neither parent should be strongly chromogenic. Another procedure has been devised for separating hybrids from their parents. The parent strains may be selected without regard to the amount of pigment they produce, but they must differ from each other in the assimilation of two carbon sources. Because this study has resulted in the first ascosporogenous yeasts known to ferment both maltose and lactose, and because the hybrids reported here may be of particular interest to geneticists, the results of this study will be presented in greater detail than was done in the previous study.

Phylogeny of the genus Kluyveromyces inferred from the mitochondrial cytochrome-c oxidase II gene.

A phylogenetic analysis of 17 species belonging to the genus Kluyveromyces and 12 reference and outgroup species was performed using mitochondrial cytochrome-c oxidase II gene sequences. The genus Kluyveromyces appears as a polyphyletic taxon formed by species included within the following four main groups. The Kluyveromyces phaffii group encompasses the species Kluyveromyces blattae, K. phaffi...

Purification and properties of a yeast nucleotide pyrophosphatase.

A nucleotide pyrophosphatase (EC 3.6.1.9) has been purified from extracts of the hybrid yeast, Saccharomyces fragilis X Saccharomyces dobzhanskii, to an extent where it appears homogeneous by ultracentrifugation and sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Multiple protein components seen on polyacrylamide and starch gel electrophoresis also exhibit enzyme activity. Ultracentr...

Reactions of Saccharomyces cerevisiae and Zygosaccharomyces bailii to sulphite.

Sulphite inhibited growth of all four yeasts studied, Zygosaccharomyces bailii NCYC 563 being most sensitive and Saccharomyces cerevisiae NCYC 431 the least. Vertical Woolf-Eadie plots were obtained for initial velocities of 35S accumulation by all four yeasts suspended in high concentrations of sulphite. Equilibrium levels of 35S accumulation were reached somewhat faster with strains of S. cer...

Genome sequence of the highly weak-acid-tolerant Zygosaccharomyces bailii IST302, amenable to genetic manipulations and physiological studies

Zygosaccharomyces bailii is one of the most problematic spoilage yeast species found in the food and beverage industry particularly in acidic products, due to its exceptional resistance to weak acid stress. This article describes the annotation of the genome sequence of Z. bailii IST302, a strain recently proven to be amenable to genetic manipulations and physiological studies. The work was bas...

Construction of High Ethanol Fermenting Halotolerant Hybrid by Intergeneric Protoplast Fusion of Saccharomyces cerevisiae and Zygosaccharomyces rouxii

Saccharomyces cerevisiae M30, a high ethanol producing flocculent strain, was hybridized with Zygosaccharomyces rouxii TISTR1750, a halotolerant non-flocculent strain, by polyethylene glycol induced protoplast fusion. The protoplast fusion of these two yeast genera occurred with frequency of 5.89X10-5. Three fusants, namely RM11, RM37 and RM58, were selected based on their high ethanol fermenta...
